I have central air conditioning on my second floor about 4years old and there is one room on the floor where the cold air does not blow. It is freezing in the other rooms but that one room it hardly blows out of the vent? what should I be checking for? as far I remember it always was like that even when I had the central air installed.

I used to install HVAC. This sounds like a there may be a damper in the ductwork or on the plenum that needs adjustment ( turning it by hand).

Or it could be a disconnected duct
crushed duct
etc.

hopefully you can access an attic area where the ductwork is ? If so you should be able to spot something obviously wrong. I think a damper could adjusted to cut of this room, that would be easy to fix.

something that looks like this....

You want that handle in line ( horizontal to the duct) to be 100% open. Perpendicular closes it. This one is mostly closed.

In the pic above...if its there you see the duct where it attaches to the A/C. If that pipe goes directly to that room there should be a damper a few inches from where the duct is attached. You can open or close it to increase the airflow. If you do have that flexible duct ck for sharp bends like the 1 in the pic and air leaks also. If that pipe off the A/C goes to more than 1 room then the damper may be in the Y where the duct splits to go to the different rooms. If the duct goes in walls, between floors , etc that is where you would most likely find kinks and blockage.
If you have hard pipe instead of the flex junk the dampers should be located in the same spots. Ive also seen people throw something in the attic on top of the duct and squish it enough to block off the air.

Where is the air handler ? I mean the furnace not the condenser ( assuming a heat pump)

on the roof, in a closet on the 2nd floor, 1st floor, etc.

are the supply vents in the ceiling, floor, wall ?

If the FAU ( forced air unit) aka air handler, furnace is in a 2nd floor closet and the vents are cut into the ceiling then there has to be attic area the way I see it. This is good since there has to be a way to access this area. Look for about a 2' by 3' access panel ( usually made of sheetrock), and it will be framed in. Look in bedroom closets. You of course will need a 6' stepladder.

I assume this building has a normal peaked roof.

Once you get in the attic it will be hot and nasty, could have many different kinds of insulation. Some of the more modern stuff is really nasty, but better than pink fiberglass rolled stuff. Be careful you don't want to fall through a ceiling. Take something to span the trusses to give yourself something to walk on like some scrap lumber.

Maybe you can find an HVAC guy for cheap on CL ?

bedroom closet is a good place to start.

You looking for something that is at least 4-6 SF, not like your looking for a lightswitch.

I'm sure the building code requires access.

It could also be too close to the return air ( the one that sucks air in to be re-circulated), no easy fix for that.

The register might also be installed backwards and be blowing the air in the wrong direction. They are usually held by two 1/4" hex head screws.

Some sort of deflector shield thingamajig is a possibility.

At this point I'd look at flipping the register 180 degrees. A 1/4 " chuck ( a few dollars) that fits into a drill is what you need to try this fix.
